EGFP (enhanced green fluorescent protein) mRNA can express an enhanced green fluorescence when transfected into cells, it is a direct-detection reporter mRNA with fluorescence emission at 509 nm, used as control to study transfection and expression in mammalian cells by various fluorescence-based assays. Our ARCA EGFP mRNA (5-moUTP) is provided at a concentration of 1mg/ml, it is ARCA capped, polyA-tailed and incorporated with modified nucleotides. Anti-Reverse Cap Analog (ARCA) is a cap analog used during in vitro transcription for the generation of capped transcripts. ARCA obtains the ability only inserting in the proper orientation, results in forming mRNAs that can be translated twice efficiently as those initiated with mCAP. The capping of ARCA ensures high translation efficacy, while the addition of 5-moUTP (5-Methoxy-UTP) and poly(A) tail reduces the host cell immune response and the toxicity to the modified mRNA , thus protects viable mRNA constructs. Polyadenylation (i.e. addition of Poly(A) tail) also plays an important role in the stabilization of RNA in eukaryotes and enhances the efficiency of translation initiation.
