-20°CNocodazole, an inhibitor of microtubule formation, was found to greatly inhibit cell division around the mound stage as well as during the vegetative growth phase, when applied to exponentially growing Ax-2 cells. Mitotic arrest of cells with nocodazole can be used to study phosphorylation of microtubule-associated proteins which increase during mitosis, concomitant with increased microtubule dynamics. Nocodazole has been found to dramatically inhibit T cell antigen receptor (TCR) signaling, independently of its ability to depolymerize microtubules. Nocodazole arrests the cell cycle at G2/M phase and prevents phosphorylation of the T cell antigen receptor and inhibits its activity.||Synonyms: |Methyl N-(5-thenoyl-2-benzimidazolyl)carbamate; Methyl [5-(2-thienylcarbonyl)-1H-benzimidazol-2-yl]carbamate; Oncodazole; R 17934; [5-(2-Thienylcarbonyl)-1H-benzimidazol-2-yl]-carbamic acid methyl ester||CAS No: |31430-18-9||Molecular Formula: |C14H11N3O3S||Molecular Weight:|301.32||Appearance: |Supplied as a light orange to brown powder||Purity: |≥98% (HPLC)||Melting Point: |295-305ºC||Method for Determining Identity:|Proton NMR Spectroscopic Analysis||Solvent Solubility (g/100ml):|DMSO: 1 (with warming)|Formic Acid: 15.30|Acetic Acid: 1.39|Lactic Acid: 1.79|PEG 400: 0.80|Methanol: 0.026|NaOH, 0.1N, pH 12.3: 0.015|Ethanol: 0.011|Water: < 0.0005|HCl, 0.1N, pH 1.2: 0.007|Citrate-Phosphate Buffer, pH 8.0: 0.0001|Note: Nocodozole is relatively insoluble. ||Note: Nocodozole is relatively insoluble. ||Usage:|Prepare stock solution at 5mg/ml in DMSO. Further dilutions can be made in culture media. Due to insolubility (nocodazole is only slightly soluble in water), no more than 10ug/ml concentration can be used in cell culture systems. Nocodazole is stable in biological media for seven days.||Storage and Stability:|May be stored at RT for short-term only. Long-term storage is recommended at -20°C for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.||Important Note: |This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. ||Toxicity and Hazards: |All products should be handled by qualified personnel only, trained in laboratory procedures.
