ELISA Kit Principle | Aviva Systems Biology 17-Hydroxyprogesterone ELISA Kit (OKEH02534) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-17-Hydroxyprogesterone antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated 17-Hydroxyprogesterone and incubated. The 17-Hydroxyprogesterone found in the sample or standards competes with the biotinylated 17-Hydroxyprogesterone for limited binding sites on the immobilized anti-17-Hydroxyprogesterone antibody. Excess unbound biotinylated 17-Hydroxyprogesterone and sample or standard 17-Hydroxyprogesterone is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated 17-Hydroxyprogesterone captured in the well and inversely proportional to the amount of 17-Hydroxyprogesterone which was contained in the sample or standard. | ||||||||||||||||||||||

Description of Target | 17-Hydroxyprogesterone (17-OH progesterone or 17OHP) is a C-21 steroid hormone produced during the synthesis of glucocorticoids and sex steroids. As a hormone, 17OHP also interacts with the progesterone receptor. Measurements of levels of 17-hydroxyprogesterone are useful in the evaluation of patients with suspected congenital adrenal hyperplasia as the typical enzymes that are defective, namely 21-hydroxylase and 11beta-hydroxylase, lead to a build-up of 17OHP. In contrast, the rare patient with 17alpha-hydroxylase deficiency will have very low or undetectable levels of 17OHP. 17OHP levels can also be used to measure contribution of progestational activity of the corpus luteum during pregnancy as progesterone but not 17OHP is also contributed by the placenta. |
