It is located predominately in dendrites where it accelerates tubulin polymerization, cross-links microtubule fibers into stable bundles, and connects microtubule filaments to intermediate filaments. Alternative splicing of MAP2 mRNA gives rise to at least three different isoforms including; MAP2a (280kD), MAP2b (200kD), and MAP2c (70kD). The MAP2c isoform is expressed in embryos, whereas the MAP2a and MAP2b isoforms are expressed in adult tissues. MAP2 is the somatodentritic MAP of neurons, but it also is expressed to a lesser extent in glia.||Applications: |Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.||Recommended Dilution:|Western Blot: detects MAP2 in 20ug of rat brain microsomal protein preparation. Rat brain microsomal preparation was resolved by electrophoresis, transferred to nitrocellulose and probed with M2350-02 (0.5ug/ml). Proteins were visualized using an IgG (HRP) goat anti-mouse and a chemiluminescence detection system.|Immunohistochemistry: detects MAP2 in rat brain sections.|Optimal dilutions to be determined by the researcher.||Recommended secondary antibodies:|I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo|I1904-06H IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo|I1904-06F: IgG, H&L (FITC) (X-Adsorbed) Pab Gt xMo|I1904-23: IgG, X-Adsorbed (DTAF) Pab Gt xMo||Storage and Stability:|Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. ||Note: Applications are based on unconjugated antibody.
