-20°CProliferating cell nuclear antigen (PCNA), formerly known as cyclin, is a 36kD intranuclear acidic protein closely associated with proliferating cells. PCNA was originally identified in patients with systemic lupus erythematosus. It represents a component of DNA polymerase-b. The presence of PCNA is closely associated with the cell cycle. PCNA is expressed in high concentration in the late G1-phase before the DNA synthesis, and it reaches a maximum concentration in the S-phase. The level of PCNA declines during the G2 and M phases. ||Applications:|Suitable for use in Flow Cytometry, Immunohistochemistry, Immunoprecipitation and Western Blot. Other applications not tested. ||Recommended Dilutions: |Immunohistochemistry: 1:50-1:100 (Paraffin). Requires membrane permeabilization. Does not require protein digestion pre-treatment or antigen retrieval of paraffin sections. |Immunohistochemistry: Frozen sections|Flow Cytometry: 1:100 Use 10ul to label 10e6 cells in 100ul. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. |Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. ||Note: Applications are based on unconjugated antibody.
