ELISA Kit Principle | Aviva Systems Biology General Adenosine triphosphate ELISA Kit (OKEH02624) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-Adenosine triphosphate antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated Adenosine triphosphate and incubated. The Adenosine triphosphate found in the sample or standards competes with the biotinylated Adenosine triphosphate for limited binding sites on the immobilized anti-Adenosine triphosphate antibody. Excess unbound biotinylated Adenosine triphosphate and sample or standard Adenosine triphosphate is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated Adenosine triphosphate captured in the well and inversely proportional to the amount of Adenosine triphosphate which was contained in the sample or standard. | ||||||||||||||||||||||

Description of Target | Adenosine-5'-triphosphate (ATP) is a multifunctional nucleoside triphosphate used in cells as a coenzyme. It is often called the "molecular unit of currency" of intracellular energy transfer. ATP transports chemical energy within cells for metabolism. It is one of the end products of photophosphorylation and cellular respiration and used by enzymes and structural proteins in many cellular processes, including biosynthetic reactions, motility, and cell division. One molecule of ATP contains three phosphate groups, and it is produced by ATP synthase from inorganic phosphate and adenosine diphosphate (ADP) or adenosine monophosphate (AMP). The three main ways of ATP synthesis is substrate level phosphorylation and oxidative phosphorylation in cellular respiration, and photophosphorylation in photosynthesis. |
