This MAb reacts with a 58kD protein identified as vimentin. It shows no cross-reaction with other closely related intermediate filament proteins (IFP’s) such as desmin, keratin, neurofilament, and glial fibrillary acid protein. Anti-vimentin alone is of limited value as a diagnostic tool; however, when used in panels with other antibodies, it is useful for the sub-classification of a given tumor. Expression of vimentin, when used in conjunction with anti-keratin, is helpful when distinguishing melanomas from undifferentiated carcinomas and large cell lymphomas. All melanomas and Schwannomas react strongly with anti-vimentin. It labels a variety of mesenchymal cells, including melanocytes, lymphocytes, endothelial cells, and fibroblasts. Non-reactivity of anti-vimentin is often considered more useful than its positive reactivity, since there are a few tumors that do not contain vimentin, e.g. hepatoma and seminoma. Anti-vimentin is also useful as a tissue process control reagent.||Applications: |Suitable for use in ELISA, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.||Recommended Dilution:|Western Blot: 0.5-1ug/ml|Immunoprecipitation: 1-2ug/500ug protein lysate|Immunohistochemistry (Frozen & Formalin-fixed): 0.5-1ug/ml for 30 minutes at RT|(Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes)|Optimal dilution for a specific application should be determined.||Positive Control:|Jurkat cells, Sarcomas, Melanomas||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. |For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.||Note: Applications are based on unconjugated antibody.
