ELISA Kit Principle | Aviva Systems Biology Insulin ELISA Kit (Sheep) (OKCA00326) is based on standard sandwich enzyme-linked immuno-sorbent assay technology. A Sheep monoclonal antibody specific for Sheep Insulin has been pre-coated onto a 96-well plate (12 x 8 Well Strips). Standards or test samples are added to the wells and incubated. After washing, a biotinylated detector antibody specific for Sheep Insulin is added, incubated and followed by washing. Avidin-Biotin-Peroxidase Complex is then added, incubated and unbound conjugate is washed away. An enzymatic reaction is produced through the addition of TMB substrate which is catalyzed by HRP to generating a blue color product that changes to yellow after adding acidic stop solution. The density of yellow coloration read by absorbance at 450 nm and is quantitatively proportional to the amount of sample Sheep Insulin captured in well. | ||||||||||||||||||||||||||||||||||||||||||

Description of Target | After removal of the precursor signal peptide, proinsulin is post-translationally cleaved into two chains (peptide A and peptide B) that are covalently linked via two disulfide bonds. Binding of this mature form of insulin to the insulin receptor (INSR) stimulates glucose uptake. A variety of mutant alleles with changes in the coding region have been identified. There is a read-through gene, INS-IGF2, which overlaps with this gene at the 5 region and with the IGF2 gene at the 3 region. |
