Arrays for the study of glycosylation - Glycome


Glycome is the set of sugars, free or present in more complex molecules, of an organism.
Glycome Arrays allow researchers to efficiently obtain glycosylation profiles in their samples. The capture antibodies are printed on glass slides and the glycans of these capture antibodies are removed. The glass slide arrays are pre-blocked and ready to be incubated with the samples. After incubation with the samples and washing to remove unbound proteins, five unique biotin-labeled lectins are incubated with the matrix to allow binding of the captured target proteins to the glycans. A streptavidin-conjugated fluorescent dye (Cy3 equivalent) is then added to the matrix. Finally, the glass slide is dried and a laser fluorescence scan is used to visualize the signals. These signals are then compared to the array map to identify the glycosylated proteins present in the samples.
The glycosylation profiles of several hundred human proteins can be detected simultaneously, including cytokines, chemokines, adipokines, growth factors, angiogenic factors, proteases, soluble receptors, soluble adhesion molecules in cell culture supernatants, serum, plasma and other types of samples.
