Assay Principle: 

The DetectX® Myeloperoxidase (MPO) Human ELISA Kit quantitatively measures MPO levels in serum, platelet-poor heparin plasma, saliva, urine, and tissue culture media. This sandwich ELISA has a run time of 2.5 hours. Please read the complete kit insert for more information before performing this assay. Use our provided MPO standard to generate a standard curve for the assay.

Protocol Summary:

  • Introduce standards or diluted samples into the included transparent microtiter plate, pre-coated with anti-human MPO monoclonal antibody.
  • Incubate the plate covered, shaking for 1 hour at room temperature.
  • Post-incubation, wash the plate and add the peroxidase-conjugated human MPO antibody.
  • Incubate again, covered and shaking, for 1 hour at room temperature. The immunological reaction inversely correlates with the MPO concentration in the sample.
  • Wash away excess conjugate and add the TMB substrate, which reacts with the bound antibody to produce a detectable signal.
  • Measure the signal at 450nm using a plate reader and calculate MPO concentration based on the standard curve.

Background:

Myeloperoxidase (MPO) is a tetrameric heme-containing protein abundantly produced in neutrophil granulocytes, where it plays a critical anti-microbial role. Stored in azurophilic granules, MPO is released during neutrophil degranulation and contributes to the formation of hypochlorous acid from hydrogen peroxide and chloride ions in the ‘respiratory burst.’ It also catalyzes the oxidation of tyrosine to tyrosyl radicals, both of which are cytotoxic agents against bacteria and other pathogens. A hereditary MPO deficiency can lead to increased susceptibility to infections.

MPO levels have significant clinical implications, especially in cardiology. Numerous studies have established a correlation between elevated MPO levels and coronary artery diseases, positioning MPO as a potential predictor for myocardial infarction in patients with chest pain. The combination of MPO and C-reactive protein (CRP) measurements has been found to provide a more accurate assessment of mortality risk than CRP alone, highlighting the relevance of MPO in diagnostic and prognostic healthcare settings.
