Insect cell culture medium
The Insect cell culture medium plays an essential role in providing an optimal environment for cell growth and maintenance. Commonly used media, such as Grace or Schneider, are used as a base. They are supplemented with fetal bovine serum (FBS) at concentrations of 5 to 10%, ensuring the supply of essential nutrients and growth factors crucial for cell vitality. To counter potential bacterial and fungal contamination, antibiotics and antimycotics, including penicillin-streptomycin, are added to the medium. L-glutamine is introduced to promote cell proliferation, while heparin, if required, helps prevent cell aggregation.
Additional components include glucose or dextrose as a source of carbon and energy, maintaining cellular metabolic activity. Precise pH regulation is achieved by adding sodium bicarbonate or a CO2 buffer. Essential mineral salts are incorporated to meet the nutritional needs of the cells. Depending on the specific insect cell line, growth factors can be added to optimise cell growth and functionality. Incubation is carried out at temperatures of around 27-28°C, corresponding to the preferred conditions for insect cell cultures.
