The G34W mutation of histone H3.3 has recently been identified as a reliable marker for benign and malignant bone tumor like Giant Cell Tumor (GCTB).
A study published in August 2017 uses RevMab's anti-H3.3 G34W rabbit monoclonal antibody (Clone RM263) which we market.
In this study, the authors investigated the sensitivity and specificity of anti-histone H3.3 G34W rabbit monoclonal antibody (clone RM263; RevMAb Biosciences) in a wide variety of tumours, including histological imitations of GCTB to assess its value as a diagnostic marker. The study focused on the genotyping and immunomarking of a series of osteosarcomas to determine the incidence of H3.3 G34 mutations in these primary malignant bone tumors.
This study demonstrates that anti-histone H3.3 G34W rabbit monoclonal antibody (clone RM263) is highly specific and sensitive for tumors hosting this mutation and is a potent marker of GCTB, significantly improving diagnostic accuracy in biopsy samples. Histone H3.3 G34W expression is observed exclusively in the mononuclear cell population, confirming that the neoplastic cell population is represented by mononuclear cells and not by giant osteoclast cells.
This antibody recognizes only the mutated G34W form of histone H3.3 and not the native form.
