Storage temperature
Store qPCR Green Master Mix (2x) at -30 to -15 °C in a constant temperature freezer. Minimize freeze-thaw cycles by storing in working aliquots. Protect from light.

Quality control assays
- Genomic DNA contamination:
qPCR Green Master Mix is screened for the presence of bacterial genomic DNA. Trace amounts of E. coli gDNA may be detected through a highly sensitive real-time PCR assay using primers specific for the E. coli 16S rRNA gene. Acceptable limits are established according to the quantity of DNA contaminant to an amount not enough to originate a positive signal.
- Nuclease assays:
To test for DNase contamination, 0.2-0.3 μg of pNZY28 plasmid DNA are incubated with the master mix for 14-16 h at 37 °C. To test for RNase contamination, 1 μg of RNA is incubated with the master mix for 1 h at 37 °C. Following incubation, the nucleic acids are visualized on a GreenSafestained agarose gel. There must be no visible nicking or cutting of the nucleic acids.
- Functional assay :
qPCR Green Master Mixes are extensively tested for activity, processivity, efficiency, sensitivity and heat activation.
