Datasheet SDS WB Stripping Buffer is used for recycling deproteinized membranes in western blot (WB). After carrying out a WB, a further WB may be need to be carried out with proteins with stable expression levels like actin and tubulin for reference, or to test other proteins for comparison. This reagent removes primary and secondary antibodies from the membrane by dissolving the conugation between primary and secondary antibodies and antigens, allowing the same membrane to be reused to detect other proteins. Compared with the SDS-PAGE gel, this reagent helps to eliminate the loading error and makes the experiment more comparable and accurate. This reagent is non-toxic, is free of β-Mercaptoethanol, and can be used at room temperature. This reagent can be used for at least 5 WB detections on the same membrane, and the whole detection can be completed within 15-30 min. This product is efficient and and does not affect proteins.

Directions for use | Protocol: 1. Wash the membrane with TBS-T 3 times, 10 min each time. Immerse the membrane into an appropriate volume of WB Stripping Buffer, incubate at room temperature for 15-30 min and shake slowly. (Note: For some antibodies, a washing time of 30-60 min may be required).2. Using tweezers, remove the membrane. Elute with TBS-T once and wash for 5 min. 3. Seal with skimmed milk powder or BSA. Notes: 1. For horseradish peroxidase systems, 5% skimmed milk powder should be used for each seal. For alkaline phosphatase systems, casein should be used for each seal.2. PVDF membrane is recommended with this product. A high amount of protein loss has been observed when using this WB stripping buffer with NC membrane. 3. This buffer can be used for WB detection with ECL similar chemiluminescence reagents. This buffer is not compatible with non-chemiluminescence reagents, such as DAB, NBT/BCIP. 4. Please wear personal protective equipment, such as lab coats, safety glasses and disposable gloves, when using this product. |
