Le MG-132 [Z-Leu- Leu-Leu-CHO] est un inhibiteur du protéasome puissant, réversible et perméable aux cellules. Le MG-132 [Z-Leu- Leu-Leu-CHO] est utilisé comme outil pour perturber la dégradation des protéines intracellulaires régulée par le protéasome. Ces protéines traitées par le protéasome sont impliquées dans les processus inflammatoires et la régulation du cycle cellulaire, ce qui confère une grande pertinence aux inhibiteurs aldéhydiques du protéasome de ce type. Par exemple, le MG-132 [Z-Leu- Leu-Leu-CHO] supprime l'activation de NF-kappaB (à 10 muM) en empêchant la dégradation d'IkappaB (IC50=3 muM), ce qui empêche le facteur de transcription de fournir une chimiorésistance aux cellules cancéreuses. Il a également été démontré que le MG-132 [Z-Leu- Leu-Leu-CHO] empêche le clivage de la protéine précurseur de l'amyloïde (APP) par la bêta-sécrétase. De plus, le MG-132 [Z-Leu- Leu-Leu-CHO] est un inhibiteur du protéasome 26S et de la calpaïne, ainsi qu'un activateur de JNK1, c-Jun et c-Fos.Les produits apparentés incluent sc-351846 (sc-351846).
