Application Details
ELISA
Useful in Sandwich ELISA for Quantitative Detection of Antigen. Aliquot 0.1ml per well of the 50ng/ml, 25ng/ml, 12.5ng/ml, 6.25ng/ml, 3.12ng/ml, 1.56ng/ml, 0.78ng/ml mouse Chemerin standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffer into the control well (Zero well). Add 0.1ml of each properly diluted sample of mouse cell culture supernates, serum or plasma(heparin, EDTA) to each empty well. We recommend that each mouse Chemerin standard solution and each sample is measured in duplicate.

Store vials at 4°C prior to opening. Centrifuge product if not completely clear after standing at room temperature. This product is stable for 6 months at 4°C as an undiluted liquid. Dilute only prior to immediate use. For extended storage freeze at -20°C or below for 12 months. Avoid cycles of freezing and thawing.

Background
Chemerin, also known as RARRES2 or TIG2, is a protein that is encoded by the RARRES2 gene. It is mapped to 7q36.1. Chemerin is a potent chemoattractant specific for antigen-presenting cells that requires proteolytic activation and acts as a ligand for the G protein-coupled receptor CMKLR1(also known as ChemR23). It is a 14 kDa protein secreted in an inactive form as prochemerin and is activated through cleavage of the C-terminus by inflammatory and coagulation serine proteases. Chemerin was found to stimulate chemotaxis of dendritic cells and macrophages to the site of inflammation. What’s more, the active protein has several roles, including that as an adipokine, and is truncated on both termini from the proprotein.
