The TACE Activity Assay Kit is a homogeneous assay that can be used to detect the activity of enzyme and for screening of TACE inhibitors. It contains a 5-FAM FRET substrate, derived from a sequence surrounding the cleavage site of TACE. In the intact FRET peptide, the fluorescence of 5-FAM is quenched by a TACE Inhibitor. Active TACE cleaves FRET substrate into two separate fragments resulting in an increase of 5-FAM fluorescence which can be monitored at excitation/emission = 490nm/520nm. The long wavelength fluorescence of 5-FAM is less interfered by the autofluorescence of cell components and test compounds.||Kit Components:|T0500-12A: TACE substrate (4mM), 1x50ul|T0500-12B: Fluoresence Standard (5-FAM), 1mM, 1x10ul|T0500-12C: Assay Buffer, 1x20ml|T0500-12D: TACE Inhibitor (1mM), 1x10ul|T0500-12E: Stop Solution, 1x10ml||Storage and Stability: |Store all kit components at -20°C. Protect T0500-12A and T0500-12B from light and from moisture. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
