a-catenin is a 102kD protein which is related to the cytoskeletal protein vinculin. Vinculin associates with both cell-cell and cell-extracellular matrix adherens-type junctions, and has been shown to interact with both a-actinin and actin. The homology between a-catenin and vinculin is localized to three major regions (amino-terminal, central, and carboxy-terminal portions) with the c-termini exhibiting the highest overall homology . Interestingly, it is the carboxy terminal domain of vinculin which is responsible for its self-association properties . The homology between the c-termini of catenin and vinculin led to speculation that a-catenin can self-associate and/or co-associate with vinculin thereby linking the cadherin/catenin complex to the cytoskeleton. However, recent studies indicate that cadherin/catenin complexes are linked to the actin cytoskeleton via a direct association between a-actinin and a-catenin. In addition, the a-catenin protein has been found to posses both actin binding and bundling activities.||Applications: |Suitable for use in Immunofluorescence, ELISA, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested.||Recommended Dilution:|Immunofluorescence: 2-5ug/ml|ELISA: 0.1-1ug/ml |Western Blot: 1ug/ml |Immunohistochemistry ( formalin-fixed, paraffin-embedded tissue): 2-10ug/ml. Heat-Induced Epitope Retrieval (HIER) is required.
