ELISA Kit Principle | Aviva Systems Biology DHEA ELISA Kit (OKEH02530) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-DHEA antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated DHEA and incubated. The DHEA found in the sample or standards competes with the biotinylated DHEA for limited binding sites on the immobilized anti-DHEA antibody. Excess unbound biotinylated DHEA and sample or standard DHEA is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated DHEA captured in the well and inversely proportional to the amount of DHEA which was contained in the sample or standard. | ||||||||||||||||||||||

Description of Target | 5-Dehydroepiandrosterone (5-DHEA) is a 19-carbon endogenous steroid hormone. It is the major secretory steroidal product of the adrenal glands and is also produced by the gonads and the brain. DHEA is the most abundant circulating steroid in humans. DHEA has been implicated in a broad range of biological effects in humans and other mammals. It acts on the androgen receptor both directly and through its metabolites, which include androstenediol and androstenedione, which can undergo further conversion to produce the androgen testosterone and the estrogens, including estrone, estradiol, and estriol. DHEA is also a potent sigma-1 agonist. It is considered a neurosteroid. |
