-20°CMast cells contain a number of preformed chemical mediators such as histamine, chymase, carboxypeptidase and proteolytic tryptase. A substantial quantity of tryptase is estimated to be found in mast cells of skin and lung and suggestes this enzyme plays a major role in mast cell mediated events. In vitro studies indicate tryptase can cleave C3 to form C3a anaphylatoxin, inactivate fibrinogen as a coaguable substrate for thrombin and activate latent collagenase. Human mast cell tryptase has considerable potential as a marker for mast cell activation as well as being a mediator of inflammation. ||Applications: |Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.||Recommended Dilution: |Western Blot: 1:50 for 2hrs at RT.|Immunohistochemistry (Formalin/paraffin): 1:2000 for 30 min at RT. Staining of formalin/paraffin tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min.|Optimal dilutions to be determined by the researcher.||Hybridoma: |Produced by fusion between BALB/c splenocytes and myeloma NS1 cells.||Positive Control: |Tonsil||Storage and Stability:|May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
