Sequencing by synthesis consists of the addition of a fluorescent nucleotide as each dNTP is added and then cleaved to allow the incorporation of the next base.
As sequencing is performed on DNA fragments, obtained from a sample and representing a library, each fragment ends with a fluorescent base. The end result is true base-by-base sequencing that provides accurate data for a wide range of applications.
