MMPs are proteolytic enzymes capable of degrading connective tissue components. They have a common mode of activation, a conserved amino acid sequence in the putative metal binding-active site region, and are inhibited by specific tissue inhibitors of metalloproteinases (TIMPs). MMPs and TIMPs play a significant role in regulating angiogenesis. MMP-2; also known as 72kD collagenase IV or gelatinase A is synthesized as a 631 amino acid proenzyme which is activated by cleavage of the first 80 amino acids into an active form.of 62kD. ||Applications:|Suitable for use in Inhibition of MMP-2 Activity, Western Blot and Immunohistochemistry. Other applications not tested. ||Recommended Dilution: |Inhibition of MMP-2 Activity: 2-4ug/ml|Western Blot: 0.5-1.0ug/ml for 2hrs at RT. |Immunohistochemistry (Formalin/paraffin): 2-4ug/ml for 30 min at RT. No special pretreatment is required for the staining of formalin-fixed, paraffin-embedded tissues |Optimal dilution determined by the researcher. ||Positive Control: |Conditioned, serum-free medium from (TPA-treated) HFL-1 cells. Placenta.||Storage and Stability:|May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at-20°C. Aliquots are stable for at least 12 months at-20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.||Note: Applications are based on unconjugated antibody.
