Cellular Immunology Assays
We propose a set of in vitro efficacy tests using cell assays for immunological studies:
- Immunophenotyping and lymphocyte assays: Assays in whole blood, PBMCs and purified immune cell subsets.
- Toxicity tests-ADCC, ADCP, CDC: Antibody Dependent Cell Cytotoxicity (ADCC) and Complement Dependent Cytotoxicity (CDC) tests.
- Fc receptor binding
- Mixed lymphocyte reactions: Mixed lymphocyte reactions (MLR) are a common method for assessing the cellular immune response.
- Macrophage, Treg and NK cell assays
- Multiplex cytokine profiling
- ELISA and ELISpot: We propose tests with colorimetric, fluorescent, temporal resolution and homogeneous ELISA formats (not requiring washing). ELISpot assays are highly sensitive immunoassays that measure the frequency of cells secreting individual cytokines.
- Immune Function Assays: These assays are performed in human whole blood, purified PBMC or subsets and mouse splenocytes or whole blood. Whole blood provides a particularly useful format for pharmacodynamic dosages.
