The TBARS assay was developed for the measurement of MDA production via lipid peroxidation and is a well-established and widely used method for assessment of oxidative damage. However, as typically performed, the traditional method for TBARS analysis is often criticized for lack of specificity and sensitivity. Indeed, currently-available TBARS assays require boiling the sample with strong acid which also generates interfering chromophores.||This assay uses a room temperature TBARS method that eliminates the requirement for a high concentration of a strong acid and cuts reaction time in half. By running the assay at room temperature, low levels of interfering substances are generated. This results in more accurate, precise and reliable TBARS measurements.||Advantages of this BioAssay™ Kit:|Industry leading sensitivity (250nM) |Enhanced MDA measurement with use of proprietary reagent formulation |Reduced background signal|Shorter assay times; 15 minutes at RT|Useful with any biological, nutraceutical or food sample|Less expensive than other methods now on the market|Eliminates the requirement of boiling samples|Can use either cuvette or microplate formats||Principle of the Procedure:|This assay is based on the reaction of a chromogenic reagent, 2-thiobarbituric acid, with MDA at 25°C. One molecule of MDA reacts with 2 molecules of 2-thiobarbituric acid via a Knoevenagel-type condensation to yield a chromophore with absorbance maximum at 532nm, as shown in Figure 1.||Kit Components:|1. Indicator, 2-Thiobarbituric Acid, 2x1g|2. Acid Reagent, 10% Acid Solution in Dimethylsulfoxide, 1x40ml|3. MDA Standard, 10mM Malonaldehyde Tetrabutylammonium Salt, 1x100ul|4. Microtiter plate, 2x96 wells||Storage and Stability:|Store components at 4°C. Stable for 6 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
