-20°CLactate is generated by lactate dehydrogenase (LDH) under hypoxic or anaerobic conditions. Monitoring lactate levels is, therefore, a good indicator of the balance between tissue oxygen demand and utilization and is useful when studying cellular and animal physiology.||Simple, direct and automation-ready procedures for measuring lactate concentration are very desirable. Lactate assay kit is based on lactate dehydrogenase catalyzed oxidation of lactate, in which the formed NADH reduces a formazan (MTT) reagent. The intensity of the product color, measured at 56 nm, is proportionate to the lactate concentration in the sample.||Applications:|Direct Assays: Lactate in serum, plasma, and cell media samples.||Key Features:|Sensitive and accurate|Detection limit: |96-well plate assay: 0.05mM and linearity up to 2mM L-Lactate. |For cell culture samples containing phenol red: 0.1mM and linearity up to 1mM L-Lactate in 96-well plate assay.||Convenient: The procedure involves adding a single working reagent, and reading the optical density at time zero and at 20 minutes. Room temperature assay. No 37°C heater is needed.||High-throughput: Can be readily automated as a high-throughput 96-well plate assay for thousands of samples per day.||Kit Components: (100 tests in 96-well plates):|L1011-04A1: Assay Buffer, 1x10ml |L1011-04A2: NAD Solution, 1x1ml|L1011-04A3: Enzyme A, 1x120ul |L1011-04A4: Enzyme B, 1x120ul |L1011-04A5: MTT Solution, 1x1.5ml|L1011-04A6: Standard, 1x1ml Supplied in 20mM L-Lactate||Storage and Stability:|Store components at -20°C.
