Actin is a major component of the cytoskeleton and is present in most cell types. This MAb is highly specific to actin from smooth muscles. Its epitope lies in the first four N-terminal amino acids. This MAb does not stain cardiac or skeletal muscle; however, it does stain myofibroblasts and myoepithelial cells. This antibody could be used together with anti-muscle specific actin and myogenin in making a diagnosis of smooth muscle and skeletal muscle tumors. In most cases of rhabdomyosarcoma, this antibody yields negative results whereas anti-muscle specific actin and myogenin are positive. Leiomyosarcomas are positive only with anti-muscle specific actin and anti-smooth muscle actin and are negative with anti-myogenin.||Applications: |Suitable for use in Immunofluorescence, Flow Cytometry (Not Tested), FLISA, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.||Recommended Dilution:|Flow Cytometry (Not Tested): 0.5-1ug/million cells|Immunofluorescence: 0.5-1ug/ml|Western Blotting: 0.5-1ug/ml|Immunoprecipitation: 0.5-1ug/500ug protein lysate|Immunohistochemistry (Frozen & Formalin-fixed): 0.5-1ug/ml for 30 minutes at RT|No special pretreatment is required for the immunohistochemical staining of formalin-fixed, paraffin-embedded tissues.|Optimal dilution for a specific application should be determined.||Positive Control:|Blood vessels in all tissues, smooth muscle or leiomyosarcoma||Storage and Stability:|May be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Dilute only prior to immediate use. Stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
