-20°CRecognizes a 60kDa protein, identified as the heat shock protein 60 (hsp60). Its epitope is localized between aa 383-419 of human hsp60. A wide variety of environmental and pathophysiological stressful conditions trigger the synthesis of a family of proteins known as heat shock proteins (hsp’s), more appropriately called as stress response proteins (srp’s). hsp60 is a potential antigen in a number of autoimmune diseases. In human arthritis and in experimentally induced arthritis in animals, disease development coincides with the development of immune reactivity directed against not only bacterial hsp60, but also against its mammalian homolog. Clone LK1, unlike LK2, recognizes only the mammalian (not bacterial) hsp60 and is useful in distinguishing hsp60 from mammals and bacteria.||Applications: |Suitable for use in Immunofluorescence, Flow Cytometry (Not Tested), ELISA, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.||Recommended Dilution:|Flow Cytometry (Not Tested): 0.5-1ug/million cells|Immunofluorescence: 0.5-1ug/ml|Western Blot: 0.25-0.5ug/ml|Immunoprecipitation: 0.5-1ug/500ug protein lysate|Immunohistochemistry: Frozen only: 0.5-1ug/ml for 30 minutes at RT|Optimal dilutions to be determined by the researcher.||Positive Control:|HeLa or HepG2 cells. Synovial biopsies from patients with juvenile chronic arthritis. Synovial lining layer is strongly positive for hsp60. Breast carcinoma.||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. |Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.||Note: Applications are based on unconjugated antibody.
