-20°CMyelin-associated glycoprotein (MAG),a member of the immunoglobulin superfamily, is a membrane glycoprotein of 100kD, and is thought to be involved in the process of myelination. Axonal regeneration in the adult central nervous system (CNS) is limited by two proteins in myelin, Nogo and MAG. MAG has been implicated in the inhibition of nerve regeneration in the CNS.||Applications: |Suitable for use in FLISA, Western Blot, Immunocytochemistry and Immunohistochemistry. Useful for myelinogenesis and multiple sclerosis studies. Other applications not tested.||Recommended Dilutions:|Western Blot: non-reducing conditions only|Immunohistochemistry: Frozen and Paraffin|Immunocytochemistry: methanol fixation required. |1. Oligodendrocytes are cultivated on slides and fixed with methanol for 10 minutes at-20°C in the middle log-phase. |2. After blocking the nonspecific binding sites with 5% FCS the cells are incubated with the properly diluted M9757-50 in a humid chamber for one hour at RT.|3. Afterwards cells are washed and incubated with anti-Mouse IgG-FITC. The slides are washed and covered appropriately|Optimal dilutions to be determined by the researcher.||Positive Control:|Rat hippocampus tissue, rat brain lysate, PC12 cells||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. |Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. ||Note: Applications are based on unconjugated antibody.
