application example
1 – 0.1 ng SARS-CoV-2 SPIKE glycoprotein.
SARS-CoV-2 SPIKE glycoprotein samples were diluted in loading buffer to concentrations of 1 ng/µl, 0.1 ng/µl and 0.01 ng/µl and further denatured by boiling at 95°C for 5 min. Proteins were separated on 12% SDS-PAGE and blotted 1h to nitrocellulose (pore size of 0.45 µm), using semi-dry transfer. Blot was blocked with 2% milk in 4°C/ON with agitation. Blot was incubated in the primary antibody at a dilution of 1: 1 000 for 1h/RT with agitation in TBS-T. The antibody solution was decanted and the blot was rinsed briefly twice, then washed once for 15 min and 3 times for 5 min in TBS-T at RT with agitation. Blot was incubated in Agrisera matching secondary antibody (AS09 602, Goat anti-rabbit IgG horse radish peroxidase conjugated) diluted to 1:25 000 in for 1h/RT with agitation. The blot was washed as above and developed for 4 min with Agrisera ECLBright (AS16 ECL-N). Exposure time was 20 seconds.
