Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is a 36kD multifunctional protein whose main function is to catalyse the reversible oxidative phosphorylation of glyceraldehyde-3-phosphate, in conjunction with inorganic phosphate and nicotinamide adenine dinucleotide (NAD). This reaction is an important energy yielding step in carbohydrate metabolism.||GAPDH has also been shown to translocate to the nucleus under a variety of stressors, most of which are associated with oxidative stress, whereby it mediates cell death. A further report has shown that GAPDH binds to several proteins that are responsible for neurodegenerative diseases, such as amyloid precursor protein and Huntingtin (Hara et al. 2006).||Applications: |Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.||Recommended Dilution:|Western Blot: 1:1000-1:5000|Optimal dilutions to be determined by the researcher.||Hybridoma: |Sp2/0 myeloma cells with spleen cells from Balb/c mice.||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. |For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
