-20°CProliferating cell nuclear antigen (PCNA), formerly known as cyclin, is a 36kD intranuclear acidic protein closely associated with proliferating cells. PCNA was originally identified in patients with systemic lupus erythematosus. It represents a component of DNA polymerase-b. The presence of PCNA is closely associated with the cell cycle. PCNA is expressed in high concentration in the late G1-phase before the DNA synthesis, and it reaches a maximum concentration in the S-phase. The level of PCNA declines during the G2 and M phases. ||Applications:|Suitable for use in FImmunohistochemistry, Immunoprecipitation and Western Blot. Other applications not tested. ||Recommended Dilutions: |Immunohistochemistry: 1:50-1:100 (Paraffin). Requires membrane permeabilization. Does not require protein digestion pre-treatment or antigen retrieval of paraffin sections. |Immunohistochemistry: Frozen sections|Western Blot: 1:1000|Optimal working dilutions to be determined by researcher.||Recommended Secondary Reagents: |I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo|I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo||Recommended Positive Control Tissue: |Tonsil||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. |For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. ||Note: Applications are based on unconjugated antibody.
